Inverse Z-spectrum analysis for clean NOE and amide CEST-MRI – application to human glioma Moritz Zaiss, Johannes Windschuh, Jan-Eric Meissner, Daniel Paech, Alexander Radbruch, and Peter Bachert Dept. Medical Physics in Radiology, German Cancer Research Center (DKFZ), Heidelberg, Germany, Dept. of Neuroradiology, University of Heidelberg, Heidelberg, Germany, Section Neuro–oncologic Imaging, German Cancer Research Center (DKFZ), Heidelberg, Germany Purpose Endogenous chemical exchange saturation transfer (CEST) effects are always diluted by competing effects such as direct water saturation and semi-solid magnetization transfer (MT). This leads to unwanted T2 and MT signal contributions that dilute the observed CEST signal (spillover effect)1,2. Furthermore, all CEST effects appear also to be scaled by the T1 relaxation time of the mediating water pool2,3. As MT, T1 and T2 are also altered in tumor regions, a recently published correction algorithm yielding the apparent exchange-dependent relaxation AREX, is used to evaluate in vivo CEST effects2. This study focuses on CEST effects of amides (3.5 ppm) and Nuclear-Overhauser-mediated saturation transfer (NOE, -3.5 ppm) in human glioblastoma.

The z-spectrum from human blood at 7T

Chemical Exchange Saturation Transfer (CEST) has been used to assess healthy and pathological tissue in both animals and humans. However, the CEST signal from blood has not been fully assessed. This paper presents the CEST and nuclear Overhauser enhancement (NOE) signals detected in human blood measured via z-spectrum analysis. We assessed the effects of blood oxygenation levels, haematocrit, c...

On the transmit field inhomogeneity correction of relaxation‐compensated amide and NOE CEST effects at 7 T

High field MRI is beneficial for chemical exchange saturation transfer (CEST) in terms of high SNR, CNR, and chemical shift dispersion. These advantages may, however, be counter-balanced by the increased transmit field inhomogeneity normally associated with high field MRI. The relatively high sensitivity of the CEST contrast to B1 inhomogeneity necessitates the development of correction methods...

Inverse Z-spectrum analysis for spillover-, MT-, and T1 -corrected steady-state pulsed CEST-MRI--application to pH-weighted MRI of acute stroke.

Endogenous chemical exchange saturation transfer (CEST) effects are always diluted by competing effects, such as direct water proton saturation (spillover) and semi-solid macromolecular magnetization transfer (MT). This leads to unwanted T2 and MT signal contributions that lessen the CEST signal specificity to the underlying biochemical exchange processes. A spillover correction is of special i...

On the origins of chemical exchange saturation transfer (CEST) contrast in tumors at 9.4 T.

Chemical exchange saturation transfer (CEST) provides an indirect means to detect exchangeable protons within tissues through their effects on the water signal. Previous studies have suggested that amide proton transfer (APT) imaging, a specific form of CEST, detects endogenous amide protons with a resonance frequency offset 3.5 ppm downfield from water, and thus may be sensitive to variations ...

Imaging amide proton transfer and nuclear overhauser enhancement using chemical exchange rotation transfer (CERT).

PURPOSE This study investigates amide proton transfer (APT) and nuclear overhauser enhancement (NOE) in phantoms and 9L tumors in rat brains at 9.4 Tesla, using a recently developed method that can isolate different contributions to exchange. METHODS Chemical exchange rotation transfer (CERT) was used to quantify APT and NOEs through subtraction of signals acquired at two irradiation flip ang...
